PacBio Revio

The Revio system adds affordability, high throughput, and ease of use to a foundation of long reads, exceptional accuracy, and direct methylation detection.

With a high-density, 25 million ZMW SMRT Cell, up to 4 SMRT Cells in parallel, and 24-hour run times, the Revio system delivers 360 Gb of HiFi reads per day, equivalent to 1,300 human whole genomes per year. ​

HiFi sequencing provides small variants, structural variants, repeat expansions, methylation, and haplotype phasing from a single library and sequencing run. ​

Operating the Revio system is simple, thanks to a combined reagent and sample plate, rapid run setup, flexibility to queue runs while sequencing is in progress, and no external nitrogen supply. ​

Advanced algorithms on the Revio system deliver exceptional read accuracy plus methylation calling in every run. Its optimized file formats reduce data storage needs through quality value binning and smart read ordering.

The Revio system simplifies the HiFi sequencing experience with fewer consumables optimized for easy tracking and minimal waste. With workflow improvements, loading consumables takes under a minute at the instrument.

The polymerase kit contains optimized reagents to bind polymerase to 24 SMRTbell libraries in preparation for sequencing.

One tray contains 4 high-density SMRT Cells, each with 25 million zero-mode waveguides.

Library prep for the Revio system supports manual and automated preps with SMRTbell prep kit 3.0.

Sequencing plates are linked automatically to run designs through an NFC tag, enabling runs to be set up in less than one minute on instrument.

The Revio work deck is isolated from the four sequencing stages, leaving the system available up to 20 hours per day for loading consumables for subsequent runs.

The Revio system includes powerful NVIDIA GPUs, which provide more than 20× the compute power of the Sequel IIe system, to deliver accurate, ready-to-use data directly from the instrument.​

The GPUs provide rapid turnaround time for basecalling and HiFi read generation, directly outputting HiFi reads in BAM format.

DeepConsensus uses advanced deep learning techniques to further extend the accuracy and yield of HiFi sequencing.

Standard runs include DNA methylation status, calculated with a deep learning algorithm that processes polymerase kinetics.

All fundamental processing steps are performed on instrument, including barcode demultiplexing.

The Revio system produces the same — and better — read length, quality, and variant calling performance that have made HiFi sequencing so celebrated.

HiFi reads are tens of kilobases long, providing the ability to resolve large variants and map to difficult regions of the genome.​

When will the Revio system be available?

Now. The Revio system is available to order and has shipped to customers worldwide

What is the price per genome on the Revio system?

U.S. list price is $995 for sequencing reagents for one Revio SMRT Cell, which has an expected yield of 90 Gb, equivalent to a 30X human genome. Your local sales representative can provide detailed pricing in your currency.

Are SMRTbell libraries prepared for the Sequel IIe system compatible with Revio?

Yes. The same library prep protocols are available for the Sequel systems and the Revio system. SMRTbell libraries can be used across systems. The sequencing reagents — polymerase/binding kit, sequencing plate, and SMRT Cell tray — are specific to each system. Currently the Revio polymerase kit supports libraries ≥3 kb; support for all library sizes is coming soon.

Is the output file format the same between the Sequel IIe system and the Revio system?

Yes. The hifi_reads.bam format continues to adhere to the SAM specification. The Revio BAM format has minor differences – for data size efficiency – in base quality encoding and read sort order.
